Cette série, dans l'esprit des Experts et d'autres séries sur les méthodes d'enquête criminelle scientifique, raconte les aventures d'une experte anthropologiste, Temperance Brennan, appelée à travailler à partir de squelettes. L'héroïne est secondée par un agent du FBI, Seeley Booth, avec lequel elle entretient des rapports tantôt conflictuels, tantôt complices. Elle s'appuie également sur une équipe de trois scientifiques qui sont autant de personnages secondaires. Pour la seconde saison arrive un nouveau personnage, une femme, le docteur Saroyan, qui n'est autre que la petite amie de Booth mais qui sera surtout la supérieure hiérarchique de T.Brennan, surnommée aussi " Bones ".
